Uno de los grandes retos de la nanotecnología es la preparación de nuevos materiales funcionales cuyas dimensiones y estructuras estén controladas a nivel molecular o atómico. En química, se denominan nanotubos a estructuras tubulares (cilíndricas), cuyo diámetro es del tamaño del nanómetro.

Existen nanotubos de muchos materiales. Entre los más conocidos se encuentran los nanotubos de carbono (CNT) formados por láminas de átomos de carbono enrolladas, con propiedades físicas y químicas que abren un horizonte inmenso de aplicaciones.

Además de los nanotubos de carbono, existen nanotubos formados por péptidos cíclicos. Este tipo de nanoestructuras ha atraído una gran atención de la comunidad científica en los últimos años debido a sus importantes aplicaciones en biología, química y ciencia de materiales. Gran parte de este interés está relacionado con sus posibilidades tecnológicas como biosensores, materiales fotosensibles, agentes antimicrobianos, sistemas de transporte selectivo, componentes electrónicos moleculares y otros usos potenciales en biología, electrónica y óptica. La historia de estos nanotubos comenzó en 1974, cuando De Santis predijo la formación de estructuras tubulares por péptidos cíclicos, formados por aminoácidos alfa con estereoquímica alternada, D y L (D,L-α-CPs). Sin embargo, no fue hasta 1993 cuando consiguieron prepararse en un laboratorio gracias al grupo del profesor Ghadiri, en Scripps. Entre sus aplicaciones destacan las que implican la interacción con membranas, incluido el uso como antimicrobianos o como biomiméticos de los canales naturales. Su cavidad interna hidrofílica facilita el transporte de agua y de moléculas hidrofílicas de tamaño apropiado, como iones. Las propiedades externas del nanotubo vienen definidas por las cadenas laterales de los aminoácidos que componen el ciclopéptido, orientadas hacia el exterior del mismo.

Posteriormente, en el grupo del Prof. Juan R. Granja, en la Universidad de Santiago de Compostela, se han diseñado y sintetizado nanotubos formados por ciclopéptidos que alternan aminoácidos alfa con otro tipo de aminoácidos artificiales, tales como los gamma (α,γ-CPs) o los delta (α,δ-CPs). Este tipo de residuos no naturales, permite introducir grupos metileno hacia la cavidad de los canales, aumentando su hidrofobididad y permitiendo su funcionalización interior.

NanotubAR permite visualizar, utilizando la tecnología de Realidad Aumentada, cuatro tipos de nanotubos: un nanotubo de carbono (CNT) y tres nanotubos formados por péptidos cíclicos. En cada uno de ellos se combinan aminoácidos alfa naturales (L-triptófanos) con residuos sintéticos, tales como D-triptófanos (D,L-alfa-CPs), aminoácidos gamma (alfa, gamma-CPs) y aminoácidos delta (alfa, delta-CPs). En todos estos diseños los grupos amino y carbonilo de los aminoácidos quedan orientados perpendicularmente al plano de los péptidos cíclicos, con una orientación adecuada para establecer enlaces de hidrógeno entre las distintas unidades, y formar así una estructura tubular.
